背景 | Afamin, also known as AFM or alpha -Albumin,is an 84-88 kDa secreted monomeric glycoprotein member of theAlb/albumin family of molecules. Expressed by hepatocytes, CNSendothelial cells and osteoclasts, Afamin circulates in the blood atlow μg/mL concentrations. Afamin is known to bind and transportvitamin E, particularly under conditions where lipoprotein islimited. This is likely to be important in follicular fluid and CSF. Afamin also serves as an osteoclast-derived chemoattractant forpreosteoblasts, providing a rationale for the observation that boneformation often follows bone resorption. Mature mouse Afamin is 587amino acids (aa) in length (aa 22-608). It contains threeconsecutive albumin domains (aa 22-210, 211-403 and 404-599) thatcontain characteristic 5 or 6 intrachain disulfide bonds. Full-length mouse Afamin shares 67% aa sequence identity with humanAfamin. |

Detection of Mouse Afamin by Western Blot. Western blot shows mouse serum. PVDF membrane was probed with 0.2 µg/mL of Sheep Anti-Mouse Afamin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8119) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Afamin at approximately 87 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. |
