| 背景 | AFP ( alpha -Fetoprotein) is a 69‑73 kDa member of the ALB/AFP/VDB family of proteins. It is secreted by fetal liver and serves as a carrier molecule for phytoestrogens, heavy metals (Cu and Ni), estrogen and fatty acids. Mature mouse AFP is 587 amino acids (aa) in length. It contains three albumin domains (aa 20‑201, 208‑393 and 400‑591), plus 15 intrachain disulfide bonds. Rat AFP contains an alternate start site at Met287 that may generate an intracellular 37 kDa form. Mouse AFP also shows an analogous site at Met281 that may produce an equivalent isoform. Over aa 19‑605, mouse AFP shares 83% and 66% aa identity with rat and human AFP, respectively. |

Detection of Mouse alpha -Fetoprotein/AFP by Western Blot. Western blot shows lysates of mouse plasma and rat placenta tissue. PVDF membrane was probed with 1 µg/mL of Mouse alpha -Fetoprotein/AFP Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5369) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for alpha -Fetoprotein/AFP at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8. |
