| 背景 | Uba6-specific E2 Conjugating Enzyme 1 (Use1), also knownas Ubiquitin-conjugating Enzyme E2Z (UBE2Z), is a 38 to 43 kDa member of theUbiquitin-conjugating (E2) enzyme family that is highly expressed in humanplacenta, pancreas, spleen, and testis. Use1 has an E2 catalytic core domain that contains an active sitecysteine residue that is required for the formation of a thioester bond withUbiquitin. It functions specificallywith the UBE1L2/UBA6 Ubiquitin-activating (E1) enzyme, rather than the UBE1 E1 enzyme,for Ubiquitin conjugation. Expression isenriched in the cytoplasm, where it mediates the ubiquitination of RGS4 andRGS5 via the N-end rule proteolytic pathway. Use1 also functions withUBE1L2/UBA6 to mediate the conjugation of HLA F-associated Transcript 10(FAT10), a Ubiquitin-like protein, and is auto-FAT10ylated in response toTNF-alpha and IFN-gamma. Full-lengthhuman Use1 is 354 amino acids (aa) and shares 100% sequence identity with mouseand rat Use1. A potential smallerisoform lacks aa 1-108. |

PVDF membrane was probed with 0.5 µg/mL of Sheep Anti-Human Use1/UBE2Z Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8154) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Use1/UBE2Z at approximately 39-41 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. |
