| 背景 | Attractin (ATRN), also known as DPPT-L, is an approximately 200 kDa transmembrane glycoprotein that shows dipeptidyl peptidase activity similar to DPPIV/CD26. Attractin is involved in a variety of processes including monocyte-T cell adhesion, axon myelination, melanocyte pigment synthesis, and energy homeostasis. The extracellular region of human Attractin contains one EGF-like domain, one CUB domain, six Kelch repeats, four PSI domains, one C-type lectin domain, and two laminin EGF-like domains. Alternate splicing of human Attractin generates a secreted isoform that lacks the transmembrane and cyotplasmic regions. Attractin is transiently upregulated during T cell activation before expression switches to the 175 kDa secreted isoform which is released into the circulation. Soluble Attractin is preferentially expressed by leukocytes and differentiating neurons. It blocks neurite formation and is elevated in the CSF of astrocytoma patients. Within aa 84-1272, human Attractin shares 95% aa sequence identity with mouse and rat Attractin. |

Detection of Human Attractin by Western Blot. Western blot shows human plasma. PVDF membrane was probed with 2 µg/mL of Sheep Anti-Human Attractin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7238) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Attractin at approximately 190 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. | |
Attractin in HepG2 Human Cell Line. Attractin was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Sheep Anti-Human Attractin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7238) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red; Catalog # NL010) and counterstained with DAPI (blue).
