| 背景 | RASSF2 (Ras association domain-containing protein 2; also RASSF2A) is a 34‑36 kDa member of the RASSF family of proteins. It is widely expressed, and serves as a tumor suppressor by interacting with K-Ras in a GTP-dependent manner, and activating caspase-3 and inducing apoptosis. Human RASSF2 is 326 amino acids (aa) in length. It contains a bipartite NLS (aa 151‑167), a Ras-association domain (aa 176‑264) and a SARAH domain (aa 272‑319) that mediates homo- and heterotypic interactions. Multiple isoforms exist. One shows an insertion of 13 aa after Lys213, a second shows a three aa substitution for aa 305‑326, and a third shows a 20 aa substitution for aa 1‑96, accompanied by a three aa substitution for aa 231‑326. RASSF2 is often found down regulated in lung-derived tumor cell lines. |

Detection of Human RASSF2 by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line, Raji human Burkitts lymphoma cell line, CEM human T‑lymphoblastoid cell line, and Daudi human Burkitts lymphoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse RASSF2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5639) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for RASSF2 at approximately 40 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. | |
RASSF2 in U937 Human Cell Line. RASSF2 was detected in immersion fixed U937 human histiocytic lymphoma cell line using Goat Anti-Human/Mouse RASSF2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5639) at 5 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue).
