| 背景 | FHL2 (Four-and-a-half LIM domains protein 2; also SLIM3 and DRAL) is a 30‑32 kDa member of the four-and-a half class of the LIM domain-only protein family. It shows a tissue-specific expression pattern (myocardium; skeletal muscle; prostatic epithelium), and is noted for its strong transactivation of the androgen receptor (AR). Human FHL2 is 279 amino acids (aa) in length. It contains four distinct LIM domains (aa 40‑92; 101‑153; 162‑212; 221‑275) that all bind to the AR. There are two potential alternate start sites (Met115 and Met110) and one splice variant that shows a 99 aa substitution for aa 53‑279. Over aa 167‑279, human FHL2 is 95% and 96% aa identical to canine and mouse FHL2, respectively. |

Detection of Human FHL2 by Western Blot. Western blot shows lysates of HT1080 human fibrosarcoma cell line and MG‑63 human osteosarcoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human FHL2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4758) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for FHL2 at approximately 32 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8. |
