| 背景 | The Arrestin family consists of four members: Arrestin 1 (visual Arrestin), Arrestin 2 ( beta -Arrestin 1), Arrestin 3 ( beta -Arrestin 2), and Arrestin 4 (cone Arrestin). While visual and cone Arrestins are found almost exclusively in the retina, beta -Arrestins 1 and 2 are ubiquitously expressed, and were initially described as negative regulators of G protein-coupled receptor (GPCR) signaling. More recently, beta -Arrestins have been determined to serve as scaffolds for various signaling pathways, including the MAPK cascades activating ERK2, p38 alpha, and JNK3. These beta -Arrestin scaffolds tie together the appropriate kinases in series, forming a discreet signaling module that localizes components to specific subcellular environments and facilitates greater kinase activation. |

beta ‑Arrestin 2 in Human Brain. beta ‑Arrestin 2 was detected in immersion fixed paraffin-embedded sections of human brain (cortex) using 10 µg/mL Human beta ‑Arrestin 2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4564) overnight at 4 °C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue).
