背景 | The O-6-methylguanine-DNA-methyl transferase (MGMT) is involved in the direct repair of DNA alkylated at the O(6) position of guanine. MGMT transfers the alkyl group to its own cysteine thereby inactivating the MGMT molecule. Therefore, the number of MGMT molecules correlates directly with the cell’s ability to withstand DNA alkylation. |

Detection of Human MGMT by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line and MCF‑7 human breast cancer cell line. PVDF membrane was probed with 0.5 µg/mL of Human MGMT Antigen Affinity‑purified Polyclonal Antibody (Catalog # AF3794) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for MGMT at approximately 23 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. |
