背景 | TRAF-5 (TNF Receptor-Associated Factor 5; also RNF84) is a 60-66 kDa member of the A subfamily of the TRAF family of molecules. It is widely expressed, and serves as an adaptor molecule that links select TNFR family members with downstream signaling elements. In particular, it interacts with LTbR, TNFR2, OX40, CD27, CD40, CD30, RANK and TAJ/TROY, and induces NF kappa B activation. It forms homotrimers and heteromers with TRAF3. Human TRAF5 is 557 amino acids (aa) in length. It contains one RING-type Zn finger region (aa 45-85), two TRAF-type Zn finger regions (aa 127-181 and 182-239), a coiled-coil region (aa 237-342) and a C-terminal protein-protein interaction MATH domain (aa 403-549). There is one potential alternative start site 64 aa upstream of the standard start site. Full-length human TRAF-5 shares 80% aa sequence identity with mouse TRAF-5. |

TRAF‑5 in Human Lymph Node. TRAF‑5 was detected in immersion fixed paraffin-embedded sections of human lymph node using Sheep Anti-Human TRAF‑5 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3280) at 3 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue).
