背景	Mitogen- and stress-activated protein kinase 1 (MSK1), also known as ribosomal protein S6 kinase 5 (RPS6KA5), belongs to the AGC family of kinases. Both MSK1 and the related MSK2/RSKB have two kinase domains connected by a regulatory linker region, and are activated by the mitogen-activated protein kinases ERK1, ERK2, and p38. Nuclear MSK phosphorylates and activates a number of transcription factors, including ATF1 and CREB.

Detection of Human/Mouse MSK1 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line and TS1 mouse helper T cell line. PVDF membrane was probed with 1 µg/mL Goat Anti-Human/Mouse MSK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2518) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). For additional reference, recombinant human MSK1 and MSK2 (2 ng/lane) were included. A specific band for MSK1 was detected at approximately 95 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

MSK1 in HeLa Human Cell Line. MSK1 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Goat Anti-Human/Mouse MSK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2518) at 1.7 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue).
