背景 | The prohibitins, called PHB1 and PHB2, are highly conserved proteins that are present in multiple compartments in eukaryotic cells. PHB1 is 30kDa tumor suppressor protein involved in cell cycle control (1). PHB1 has been found in mitochondria, the nucleus and the plasma membrane, as well as extracellularly in circulation (2). In mitochondria prohibitins mainly exist as membrane-bound ring complexes and function as chaperones maintaining mitochondrial protein stability during protein synthesis and transportation (3,4). In the nucleus prohibitins interact with transcription factors such as Rb and p53 to regulate target gene transcription (2,5). Extracellular prohibitin can bind and activate C3 to enhance complement activation (6).抑制素,即PHB1和PHB2,是一种高度保守的蛋白质,存在于真核细胞的多个区域。PHB1是一个30kDa的肿瘤抑制蛋白,参与细胞周期控制(1)。线粒体、细胞核、胞质膜以及细胞外循环中都能找到PHB1(2)。线粒体中的抑制素主要以膜结合环复合物的形式存在且在蛋白合成和转运过程中作为分子伴侣维持线粒体蛋白的稳定性(3,4)。细胞核中,抑制素通过与转录因子如Rb和p53相互作用来调节靶基因的转录(2,5)。胞外抑制素能够结合和激活C3从而增强补体激活(6)。 |
