背景 | POLR3A is the largest subunit of the DNA-dependent RNA polymerase III, one of the DNA-dependent RNA polymerases that transcribe small non-coding RNAs such as the 5S rRNA, tRNAs and some miRNAs (1-3). Together with the second largest subunit, POLR3A forms the catalytic center of the polymerase (4). In addition, RNA polymerase III plays a role in the innate immune response by sensing non-self double stranded DNA. Transcription of the non-self DNA into RNA induces type I interferon production through the RIG-I pathway (5,6). Studies suggest that mutations in the POLR3A gene may be linked to hypomyelinating leukodystrophies; a group of inherited neurodegenerative disorders (7-9). |

Chromatin immunoprecipitations were performed with cross-linked chromatin from 4 x 106 HeLa cells and either 10 μl of POLR3A (D5Y2D) Rabbit mAb or 2 μl of Normal Rabbit IgG #2729 using SimpleChIP® Enzymatic Chromatin IP Kit (Magnetic Beads) #9003. The enriched DNA was quantified by real-time PCR using human L1M5 LINE element primers, SimpleChIP® Human tRNA-Leu Anti-Codon (TAG) Primers #12769, and SimpleChIP® Human α Satellite Repeat Primers #4486.
