背景 | RUNX3/AML2 is a member of the Runt family of transcription factors. RUNX3 is important for the suppression of cell proliferation in the gastric epithelium (1), neurogenesis of the dorsal root ganglia (2), and T cell differentiation (3,4). According to the research literature, RUNX3 is found to be inactivated in more than 80% of gastric cancers and other cancer types by gene silencing or protein mislocalization (1,5,6). The tumor suppressor function of RUNX3 is exerted by forming complexes with various transcription factors, such as Smads or β-Catenin/TCF4 to regulate downstream target gene transcription (7,8). RUNX3 is also involved in caspase-3-dependent apoptosis (9). RUNX3 is normally located in the nucleus, however, in many cancer cells, RUNX3 is tyrosine phosphorylated and mislocated to the cytoplasm. The mislocation of RUNX3 abolishes its tumor suppressor function and contributes to tumorigenesis (10). |
