| 描述 | Amphiphysin was originally described in chicken brain and located at the presynaptic terminal. It was also identified as the major autoantigen in Stiff-Man Syndrome (SMS), a neurological disease linked to an autoimmune response to antigens in the central nervous system. Amphiphysin interacts with several proteins important in the synaptic vesicle cycle. Amphiphysin contains a Src homology 3 (SH3) domain that binds to the C-terminal and proline-rich region of dynamin at the synapse. Microinjection of amphiphysin inhibits synaptic vesicle endocytosis. In addition, amphiphysin binds to AP-2 at the plasma membrane and to the inositol 5-phosphatase, synaptojanin. The interaction between synaptojanin and amphiphysin also appears to be mediated through amphiphysins SH3-domain. |
