| 描述 | The Notch signaling pathway is integral to the appropriate differentiation of multiple cell types during development. Disruption of this gene in mice leads to widespread cell death and embryonic lethality before 11.5 days of gestation. Notch signaling has also been implicated in numerous human disease processes, such as leukemia and the congenital syndromes, Alagille and CADASIL. The Notch pathway has been shown to be important in human hematopoiesis as well. Notch is a transmembrane receptor activated by multiple ligands, including Jagged1, Jagged2, and Delta1. The sequence of Jagged1 is partially homologous to the Serrate and Delta Notch ligands of Drosophila and all three share a domain unique to Notch ligands, the DSL domain. Jagged1, a 130 kDa protein, is widely expressed in developing human tissues, brain, and hematopoietic cells. In the adult brain strongest expression is found in the hippocampal formation and the Purkinje cells of the cerebellum. In cell culture assays, Jagged1 binding of Notch prevented muscle cell differentiation. Therefore, Jagged1 activation of Notch signaling pathways is important for the differentiation and proliferation of various tissues. |
