| 描述 | Adaptor proteins are critical for the formation of signaling complexes that relay signals from the plasma membrane to the nucleus. These proteins are involved in diverse pathways that regulate cell growth, differentiation, and death. GCIP (Grap2 Cyclin-D Interacting Protein) was identified in a yeast two-hybrid screen where the leukocyte-specifc adaptor protein Grap2 was used as bait. The sequence of GCIP is 65% homologous to mouse Maid, a negative regulator of transcription factors in mouse egg and zygote. GCIP protein contains a DNA binding HLH domain and a leuzine-zipper-like motif. GCIP mRNA is widely expressed in most tissues with higher expression in heart, muscle, leukocytes, kidney, and brain. Similar to Grap2, GCIP binds cyclin-D1 and the novel protein p29 GCIP interacting protein. Transfection of GCIP in cells reduces phosphorylation of Rb by cyclin-D-interacting protein kinase and inhibited E2F1-mediated transcriptional activity. Thus, GCIP may function as an adaptor protein that may regulate gene expression important for cell cycle regulation and immune signaling. |
