背景 | Chromatin Immunoprecipitation (ChIP) is a powerful method used to identifyregions of the genome associated with specific proteins. These associationsare crucial for vital cellular functions including gene transcription, DNA replicationand recombination, repair, segregation, chromosomal stability, cellcycle progression, and epigenetic silencing.1 The ChIP assay has been usedto study both histones and non-histone proteins, such as transcription factors,within the context of the cell.2,3,4,5 Transcription factors and other DNAbinding proteins have a weaker affinity than histones, which generally aretightly associated within the chromatin complex. To avoid dissociation of nonhistoneproteins from the chromatin binding site, it is necessary to incorporatea cross-link step. Kit has been optimized for usewith both histone and non-histone proteins.Using the QuickChIP Assay, cells are fixed with formaldehyde to cross-linknon-histone proteins to DNA. Alternatively, the cross-linking steps are notneeded for histone proteins, which are tightly bound to DNA. The chromatinare released from the nuclei and sheared into 200-1000 bp fragments. Thechromatin are then mmunoprecipitated with specific antibodies. DNAsequences cross-linked to the protein of interest co-precipitate as part of thechromatin complex. The DNA/Chromatin/Antibody complex is then isolatedusing Protein G Agarose. After reverse cross-linking, the associated DNA isreleased from the complex and is ready for analysis. |
