| 背景 | Hypoxia-inducible factor-1(HIF1) is a transcription factor involved in the regulation of oxygen homeostasis. It regulates the physiological responses to low oxygen levels (hypoxia) and the pathophysiology of heart attack, cancer, stroke and chronic lung disease. HIF-1 is a heterodimeric protein that consists of two subunits, HIF-1 alphaandHIF-1 beta (also known as ARNT1, the aryl hydrocarbon nuclear translocator). While HIF-1 beta is constitutively expressed, the expression of HIF-1 alpha is induced by oxygen concentrations below 6%. Heterodimerization is required to bind the hypoxia response element (HRE). |

HIF-1 alpha antibody (pAb) tested by ChIP-Seq. ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 5 µl of HIF-1 alpha antibody and 25 µg of chromatin from hypoxic Huh7 cells. ChIP DNA was sequenced on the Illumina HiSeq and 19 million sequence tags were mapped to identify Hif-1 alpha binding sites across the genome. The images show binding in the promoters of Hif1a target genes that regulate metabolism. | |
